Oruka Therapeutics is a clinical-stage biopharmaceutical company developing monoclonal antibodies (mAbs) for psoriasis and related inflammatory and immunology conditions. Oruka has no approved products and generates no revenue. Oruka's two lead programs target the IL-23 and IL-17 pathways — the same biological mechanisms behind the leading approved psoriasis treatments, Skyrizi and Bimzelx. Rather than pursuing novel targets, Oruka's thesis is to take validated biology and engineer meaningfully better product profiles, primarily through extended dosing intervals. ORKA-001 targets IL-23p19 (the same target as Skyrizi) and demonstrated a ~100-day human half-life in Phase 1, potentially enabling once- or twice-yearly dosing versus Skyrizi's every-12-week schedule. ORKA-002 targets both IL-17A and IL-17F (the same mechanism as Bimzelx) and showed a ~75-80 day half-life in Phase 1, also potentially supporting twice-yearly dosing versus Bimzelx's every-8-week schedule. Both programs are now in Phase 2 trials in psoriasis patients, with data expected in 2026-2027. Oruka also plans a sequential combination regimen (ORKA-021) pairing the two antibodies, and has a fourth program (ORKA-003) targeting an undisclosed pathway in inflammatory dermatology. Oruka holds exclusive, royalty-bearing licenses to its lead antibodies from Paragon Therapeutics, its discovery partner, and relies entirely on contract manufacturers. The company is funded by equity and intends to commercialize approved products independently or via partnerships.
