Tango Therapeutics is a clinical-stage precision oncology company developing targeted cancer therapies based on synthetic lethality — the idea that certain cancers carry specific genetic deletions that create exploitable vulnerabilities, allowing Tango's drugs to kill cancer cells while leaving normal cells largely unaffected. Tango's pipeline centers on cancers with homozygous deletion of the MTAP gene, one of the most common genetic alterations in cancer, occurring across at least 15 cancer types including roughly 40% of pancreatic cancers and 45% of glioblastoma. Tango's two lead candidates are MTA-cooperative PRMT5 inhibitors: vopimetostat, an oral drug for MTAP-deleted non-CNS cancers currently in Phase 1/2 trials, and TNG456, a brain-penetrant PRMT5 inhibitor targeting MTAP-deleted GBM.
