DESCRIPTION

Repare Therapeutics is a clinical-stage oncology biotech with no approved products or commercial revenue. Repare develops small molecule drugs based on synthetic lethality — the idea that cancer cells with certain genetic mutations become dependent on specific DNA repair pathways, which Repare's drugs can block to kill cancer cells while sparing healthy ones. The company uses a CRISPR-based discovery platform called SNIPRx to identify synthetic lethality targets across the genome. Repare's two active clinical programs are RP-3467, a Polθ ATPase inhibitor targeting tumors with homologous recombination deficiency including BRCA1/2-mutated cancers, and RP-1664, a PLK4 inhibitor targeting solid tumors and neuroblastoma with elevated TRIM37 expression. Two other programs — lunresertib (PKMYT1 inhibitor) and camonsertib (ATR inhibitor) — are on hold pending partnership deals. In early 2025, Repare cut roughly 75% of its workforce to narrow its focus. The company funds operations through partnerships and licensing deals with large pharma rather than product sales; its key collaboration with Bristol-Myers Squibb generated a $50M upfront payment with additional milestones and royalties. Repare is actively seeking partners for its deprioritized assets, and is entirely dependent on external capital to continue operations.
