DESCRIPTION

Boundless Bio is a clinical-stage oncology biotech developing small molecule drugs targeting extrachromosomal DNA (ecDNA) — circular DNA fragments found only in cancer cells that allow tumors to rapidly amplify oncogenes and develop resistance to existing therapies. The company's thesis is that conventional precision oncology drugs fail in oncogene-amplified cancers because they target proteins produced by amplified oncogenes rather than the underlying ecDNA biology. Boundless Bio instead targets the cellular machinery that ecDNA depends on to function, aiming to be selectively lethal to cancer cells while sparing healthy tissue. The company has no approved products and no revenue. Its lead candidate, BBI-940, is an oral small molecule degrader of a kinesin protein involved in ecDNA segregation during cell division, currently in a Phase 1 trial (KOMODO-1) in breast cancer patients.
