DESCRIPTION

Entrada Therapeutics is a clinical-stage biopharmaceutical company focused on developing genetic medicines for diseases that are difficult to treat because the relevant drug targets are inside cells. Entrada's core technology is the Endosomal Escape Vehicle (EEV), a proprietary cyclic peptide that enables drugs to enter cells and escape the endosome — the cellular compartment that normally traps and breaks down large molecules. Entrada conjugates EEV peptides to PMOs (short nucleic acid strands) to enable exon skipping, a technique that edits around defective portions of a gene so the body can produce a functional protein. The primary focus is Duchenne muscular dystrophy (DMD), a fatal genetic muscle disease affecting primarily boys, where patients cannot produce dystrophin, causing progressive muscle degeneration. Entrada has four DMD programs targeting different exon mutations, collectively addressing an estimated 11,500+ patients in the U.S. and Europe, with the lead program (exon 44) generating initial Phase 1/2 data expected in Q2 2026. Entrada is also expanding into inherited retinal diseases, naming its first ocular candidate, ENTR-801, targeting Usher syndrome type 2A. Entrada has no approved products and currently generates revenue through a partnership with Vertex Pharmaceuticals, which licensed Entrada's EEV technology to develop VX-670 for myotonic dystrophy type 1. Vertex paid $250M upfront and leads all development; Entrada retains rights to its DMD and ocular programs, which it intends to commercialize directly via a specialist sales force if approved.
