DESCRIPTION

Shattuck Labs is a clinical-stage biotech developing antibody therapies for inflammatory and immune-mediated diseases. Shattuck has no approved products or revenues; its value is entirely tied to its pipeline. The lead asset is SL-325, a monoclonal antibody targeting DR3, the sole receptor for a signaling protein called TL1A. The DR3/TL1A pathway drives inflammation in inflammatory bowel disease (IBD), specifically Crohn's Disease and ulcerative colitis. SL-325 is currently in a Phase 1 trial in healthy volunteers, with a Phase 2 trial in Crohn's patients expected to begin in Q3 2026. Shattuck's core scientific thesis is that blocking DR3 (the receptor) is superior to blocking TL1A (the ligand), which is the approach taken by several larger competitors. Shattuck argues that DR3 is stably expressed throughout the gastrointestinal tract, enabling continuous blockade, while TL1A-blocking antibodies trigger anti-drug antibody formation in over 64% of patients in competitor trials, reducing drug efficacy. Shattuck's pipeline also includes SL-425, a half-life extended version of SL-325, and a series of preclinical DR3 bispecific antibodies. Shattuck funds its operations through equity capital, outsources manufacturing to Kemwell Biopharma, and keeps headcount lean at 40 full-time employees. Shattuck's path to monetization involves either commercializing SL-325 directly or with a partner, or out-licensing programs to larger pharmaceutical companies.
