NextCure is a clinical-stage biopharmaceutical company developing antibody-drug conjugates (ADCs) to treat solid tumors, targeting patients who have not responded to existing therapies. NextCure has no approved products and generates no product revenue. An ADC combines a monoclonal antibody that binds to a specific protein on tumor cells with a cytotoxic payload, delivering cell-killing agents directly to tumors while minimizing damage to healthy tissue. NextCure's two lead programs are SIM0505 and LNCB74. SIM0505 is an ADC targeting CDH6, a protein overexpressed in ovarian, endometrial, non-small cell lung, and renal cancers, and is currently in Phase 1 trials in both the U.S. and China. NextCure licensed global rights outside of Greater China from Zaiming, paying $17M upfront with potential milestones and royalties owed to Zaiming. LNCB74 targets B7-H4, a protein expressed in breast, ovarian, and endometrial cancers, and is co-developed equally with LigaChem Biosciences, splitting both costs and future profits 50/50. LNCB74 is currently in Phase 1 trials in the U.S. NextCure also has earlier-stage oncology programs and preclinical non-oncology programs that it is seeking to partner or out-license rather than advance internally. If any candidates are approved, NextCure would sell to hospitals, cancer centers, and specialty pharmacies, with reimbursement primarily through Medicare and private insurers.
