DESCRIPTION

Bolt Biotherapeutics is a clinical-stage immuno-oncology company developing cancer treatments based on its proprietary Immune-Stimulating Antibody Conjugate (ISAC) platform. An ISAC combines a tumor-targeting antibody with an immune-stimulating payload, directing the innate immune system to recognize and destroy cancer cells. This is structurally similar to an antibody-drug conjugate (ADC), but instead of delivering a cytotoxic payload directly to tumor cells, an ISAC activates the immune system to do the killing biologically. Bolt argues this approach offers better tolerability and more durable anti-tumor responses than cytotoxic ADCs, though this remains to be demonstrated clinically. Bolt's lead program is BDC-4182, a claudin 18.2-targeting ISAC in Phase 1 trials for gastric and gastroesophageal junction cancer — which Bolt believes is the only claudin 18.2 ISAC in clinical development. All other pipeline programs are on hold pending additional funding or partnerships. Bolt has no approved drugs and generates no product revenue, funding itself through collaboration agreements and equity financing. Key partners include Genmab, which funds research and development activities in exchange for milestones and royalties, and Toray Industries, which co-develops a separate ISAC program. Bolt relies entirely on contract manufacturers for drug production and has 23 full-time employees based in Redwood City, California.
