Ascendis Pharma is a Danish biopharmaceutical company that develops and commercializes rare endocrine disease therapies built on its proprietary TransCon prodrug technology platform. TransCon works by temporarily binding an established parent drug to an inert carrier molecule via a self-cleaving linker, releasing the unmodified parent drug at a predictable, sustained rate. This enables extended dosing intervals while preserving the parent drug's original mechanism of action. Ascendis has two approved products: YORVIPATH (palopegteriparatide), a once-daily PTH replacement therapy for adults with hypoparathyroidism, and SKYTROFA (lonapegsomatropin), a once-weekly growth hormone for pediatric and adult growth hormone deficiency. SKYTROFA's key differentiator is weekly dosing versus the daily injections required by all other approved growth hormone therapies. A third product, TransCon CNP for achondroplasia in children, was under FDA priority review as of early 2026. Ascendis sells directly in the U.S. and select European countries, and uses distribution partners across 75+ additional countries, with exclusive license agreements covering Japan (Teijin) and Greater China (VISEN Pharmaceuticals). Both approved products serve patients with chronic conditions requiring lifelong therapy, creating a compounding revenue base as the patient pool grows. Beyond its own pipeline, Ascendis licenses its TransCon platform to partners — most notably Novo Nordisk for a once-monthly semaglutide program in obesity and type 2 diabetes — allowing it to monetize the platform in large markets without building commercial infrastructure. Ascendis targets at least €5B in annual product revenue by 2030 across roughly 20 indications and 30–40 countries.
