Chimerix is a clinical-stage biopharmaceutical company focused on developing imipridones — a class of drugs — as cancer therapies. Its lead asset is dordaviprone (ONC201), a targeted therapy for H3 K27M-mutant diffuse glioma, an aggressive and rare brain cancer with no approved targeted treatments. Patients with this mutation are automatically classified as Grade IV by the World Health Organization. Chimerix submitted an NDA to the FDA in December 2024 seeking accelerated approval for dordaviprone in recurrent H3 K27M-mutant diffuse glioma, which the FDA accepted for Priority Review with a target completion date of August 18, 2025. If approved, Chimerix plans a U.S. commercial launch in Q3 2025, targeting roughly 2,000 U.S. patients annually and a global market opportunity it estimates at over $750M. Beyond dordaviprone, Chimerix is developing ONC206, a second-generation imipridone with greater potency currently in Phase 1 dose escalation for CNS tumor patients, and ONC212, an early-stage imipridone in preclinical development. Chimerix has no product revenue today and is funded primarily by proceeds from the 2022 sale of TEMBEXA, an FDA-approved smallpox antiviral, to Emergent BioDefense for $238M upfront. Chimerix also holds rights to additional milestones and royalties from that sale, as well as licensing agreements for dordaviprone in Japan and China. In March 2025, Jazz Pharmaceuticals agreed to acquire Chimerix for approximately $935M, with the deal expected to close in Q2 2025.
